What is the difference between doing more pushups per set or doing more sets during pushup exercises?

Less reps, more sets will build muscle. More reps will build endurance.

The key to any strengthening exercise ( as opposed to cardiovascular exercise) is to do the exercise to as close as failure as possible. So with push-ups do as many per set as you possibly can, as opposed to many sets of a few reps.. . If you can do a lot of push-ups without reaching failure (50 or more), then find a way to make them harder, like one arm push-ups or plyometric push-ups, so that you reach failure sooner.

The general rule is...More reps, less weight gives you the "cut" look. Less reps and more weight builds mass.

I don't know how other guys do it, but I generally like to do a bunch of different kinds of exercises. So what I do, is do so many "REPS" (repititions, ie-# push ups.) then so many sit ups and so many other exercises, then I go back and start over and do the second set. On the second set, I start out and do the exact same number of push ups, sit up, and the rest. You want to adjust it so that you don't do 50 push ups the first set and then can't do 10 on the second. You want to make them evened out. The number of sets is how many times you go through the whole group of exercises.
